SOURCE/DESCRIPTION An EcoRI 4.0 kb single copy fragment insert into XgtWES Xb isolated from the chromosome. 21 flow-sorted library of Krumlauf et al (1982). POLYMORPHISM TaqI (T-J-CGA) identifies 2 invariant bands and a simple 2 allele polymorphism with a band at 8.4 and/or 5.8 kb. FREQUENCY St-ndled 10 unrelated individuals. 8.4 kb allele (Al) 0.65 5.8 kb allele (A2) 0.35 CHROMOSOMAL LOCALISATION Unique to chromosome 21 based on Southern blot data to normal human DNA and to that of a mouse-human somatic cell hybrid containing human 21 alone. MENDELIAN INHERITANCE Co-dominant segregation in three informative pedigrees (Utah Pedigrees K-1329, K-1331,and K-1333), 28 individuals. PROBE AVAILABILITY Probe is available for collaborative investigations pending signing of confidentiality agreement. OTHER COMMENTS No problems with RFLP analysis using standard digestion conditions.struction and characterization of genomic libraries from specific human chromosomes. PNAS 79:2971-2975 (1982). Development of an Alu-PCR Amplified YAC Probe Suitable for Enumeration of Chromosome 13 on Uncultured Lymphocytes and Amniocytes by Fluorescence in situ Hybridization
The main objective of the present study was to develop an efficient and reliable probe to be routinely used for detection of chromosome 13 copy numbers by interphase FISH. To achieve this, a Yeast Artificial Chromosome (YAC) containing sequences specific for human 13q12 (744D11), was cultured and the whole yeast genomic DNA was extracted. The human insert within the isolated DNA was amplified b...
متن کاملDevelopment of a probe consist of three cosmids to enumerate the chromosome 13 on uncultured lymphocytes or amniocytes using interphase FISH
Abstract Background: To produce a reliable probe suitable for aneuploidy detection of chromosome 13 on uncultured lymphocytes and amniocytes by fluorescence in situ hybridization (FISH), we used a contig of three overlapping cosmids mapped to 13q12.3. Methods: The cosmid DNA carrying the expected sequences of human chromosome 13 was isolated from host cells and labelled with biotin-11-dUTP. Th...
متن کاملO-27: Preimplantation Genetic Diagnosis in Prevention of Genetic Diseases -Diagnostic of Spinal Muscular Atrophy (SMA)
Background: Preimplantation genetic diagnosis - PGD is currently an established procedure allowing genetic research of oocyte or embryo before implantation to the uterus. Spinal muscular atrophy (SMA) is a neurodegenerative disorder, being the second most common lethal autosomal recessive disease in Caucasians, after cystic fibrosis. There are three clinically different types of which type I (W...
متن کاملChange in chromosome number associated with a double deletion in the Neurospora crassa mitochondrial chromosome.
The mitochondrial genome of Neurospora is usually found in a single covalently closed circular 62-kbp DNA molecule. We report here that the mitochondrial genome of a phenotypic revertant of a stopper mutant (stp-ruv) is contained primarily in two separate, nonoverlapping, autonomously replicating circular chromosomes. The circles, one about 21 kbp and the other somewhat less than 36 kbp are der...
متن کاملEvaluation of four DNA typing techniques in epidemiological investigations of bovine tuberculosis.
DNA fingerprinting techniques were used to type 273 isolates of Mycobacterium bovis from Australia, Canada, the Republic of Ireland, and Iran. The results of restriction fragment length polymorphism (RFLP) analysis with DNA probes from IS6110, the direct repeat (DR), and the polymorphic GC-rich sequence (PGRS) were compared with those of a new PCR-based method called spacer oligonucleotide typi...
